Default Negotiate vacation

Can I negotiate when at a job interview? Is taking a vacation just after starting a new job going to cost me the job offer? I am getting very tired, I need a break but I prefer to start a new job first and going on vacation right after.

Maybe it is good idea not to negotiate a vacation but just to tell them that you can start in 3 weeks and this way you can give your 2 week notice and go on vacation right after. If they really want to hire you they won't make a fuss about it.
